Recognizing the tolerance to higher concentration of toxic oxyaniones in these bacteria will assist to determination of their potential for bioremediation. We have isolated ten moderately halophilic spore forming bacilli from various saline soils in Iran and identified their intrinsic high-level-resistance to chromate, arsenate and 4 other toxic oxianions (potassium tellurite, sodium selenite, sodium selenate and sodium biselenite) by agar dilution method. Minimum inhibitory concentration (MIC) for each oxianion was determined, as the lowest concentration of inhibitor that preventing of growth at 34 °C after 72h incubation. All bacteria tested (10 isolates) were resistant to higher concentrations of arsenate. The strains B2 and C2 could tolerate arsenate up to 250 mM. Unusual resistance was observed toward chromate oxianions. Some isolates, the strains A2, B1 and C2 could tolerate sodium chromate up to 75 mM, while strain C2 showed tolerate to potassium chromate up to 70 mM and other strains (A1, A2, B1C1, C4) could showed resistance to chromate up to 60 mM. The resistance level of the isolates to selenooxianions was between 10-40 mM. The maximum tolerance was showed with selenite and the minimum tolerance was found with biselenite oxianions. The highest toxicity was found with tellurite (up to 3 mM). The influence of salinity on the toxicity of oxianions was also stadied. Increasing the salinity from 5% (w/v) to 15% (w/v), in general, enhanced sensitivity to toxic oxianions.}, keywords = {Chromate,Halophilic bacteria,selenite,Tellurite,tolerance}, title_fa = {ارزیابی مقاومت به اکسی آنیون های سمی در باسیل های اسپور دار نمک دوست نسبی بومی ایران و اثر شوری بر روی این مقاومت}, abstract_fa = {میکروارگانیسم‌های نمک دوست نقش مهمی را در پاکسازی زیستی ترکیبات سمی فلزی در محیط ایفا می‌کنند و شناسایی سویه‌های تحمل پذیر به این ترکیبات، مرحله نخست دراستفاده از این میکروارگانیسم‌ها در فرایند‌های پاکسازی زیستی است. دراین راستا 10 سویه باکتری تشکیل دهنده اسپور از خاک‌های شور درایران جداگشت و مقاومت ذاتی بسیار بالا نسبت به اکسی‌آنیون‌های سمی، کرومات، ارسنات، سلنیت، سلنات و بی‌سلنیت دربعضی از سویه‌ها نشان داده شد. روش رقت درآگار برای تعیین تحمل پذیری باکتری‌ها نسبت به این ترکیبات سمی مورد استفاده قرار گرفت و حداقل غلظت مهار کننده رشد (MIC) برای هر اکسی آنیون در دمای 34 درجه سانتی‌گراد و در زمان گرماگذاری 72 ساعت تعیین گشت. براساس تعیین MIC، همه باکتری‌های مورد آزمایش بالاترین مقاومت را نسبت به اکسی‌آنیون سمی ارسنات نشان دادند. سویه‌های B2 و C2 می‌توانستند تا غلظت250 میلی‌مول ارسنات را تحمل کنند. یک مقاومت غیر معمول نسبت به اکسی آنیون‌های کرومات در بعضی سویه‌ها مشاهده گشت. سویه‌های A2، B1 و C2 سدیم کرومات را تا 75 میلی مول تحمل می‌کردند درحالیکه سویه C2 تحمل به پتاسیم کرومات را تا 70 میلی‌مول نشان داد و سایر سویه‌ها (A1، A2، B1، C1 و C4 ) مقاومت را نسبت به پتاسیم کرومات را تا حداکثر 60 میلی‌مول نشان دادند. سطح مقاومت سویه‌ها به سلنواکسی آنیونها بین 10 تا 40 میلی‌مول بود. بالاترین مقاومت به سلنات و پایین‌ترین مقاومت به بی سلنیت نشان داده شد. بالاترین سمیت را در میان اکسی آنیون‌های مورد آزمایش را تلوریت نشان داد (حداکثر تحمل پذیری 3 میلی مول). اثر شوری بر روی سمیت اکسی آنیون‌ها مورد مطالعه قرار گرفت. افزایش شوری از (w/v) 5% به (w/v)15% بطورکلی سبب افزایش مقاومت به اکسی آنیونها در سویه‌ها می‌گردد}, keywords_fa = {Chromate,Halophilic bacteria,selenite,Tellurite,tolerance}, url = {https://jos.ut.ac.ir/article_16830.html}, eprint = {https://jos.ut.ac.ir/article_16830_bb90f662f055d3f7fc97faabce385935.pdf} }
