The GH3/B6 cells are generally grown in Ham's F12 medium supplemented with 15% horse serum and 2.5% fetal calf serum. In routine conditions, the doubling time of GH3/B6 cells varies from 30 to 52hr, depending on the qualities of the serum. The GH3/B6 cells are used to study the effects of several hormones on prolactin secretion. The recent works have shown that pectic acid increases secretion of prolactin. Pectic acid releases stored prolactin in cells and after a short time it appears to be effective. After being incubated with pectic acid for 30 minutes the cells started to become circular, protuberant shape and entered the secreting phase. pectic acid had not significant effect in number of cells but a decrease in total amount of protein was observed. After pectic acid was added, a significant decrease happened in the medium pH and this decrease caused unfavorable condition for cells. To solve this problem, the medium with Hepes was used. Hepes had been used extensively in cell culture due to its excellent buffering capacity in the physiological range. After a while; however, the cells appearance changed, their growth decreased, cytoplasmics vacuoles and membrane inclusion bodies developed and the viability decreased. Hepes stimulates the production of toxic oxygen metabolites such as hydrogen peroxide which itself results in a decrease in growth of GH3/B6 cells. Consequently, Hepes was replaced by NaOH in this experiment. NaOH had no effect on the cells and the pH was adjusted in 7.2.}, keywords = {GH3 B6 Cells,GH3 B6 cells دودمان سلولی,Cell Strain,Prolactin,Pectic Acid}, title_fa = {بررسی اثر اسید پکتیک بر مورفولوژی، بقا و عملکرد دودمان سلولی GH3/B6}, abstract_fa = {دودمان سلولی هیپوفیز موش صحرایی تحت عنوان GH3و زیر کلون آن GH3/B6 ویژه ترشح هورمون پرولاکتین و هورمون رشد می‌باشند. این سلولها بطور گسترده برای بررسی اثر مواد مختلف روی ترشح پرولاکتین به صورت”in vitro“ بکار می‌روند. محیط کشت مـورد استفاده برای این سلولها، Ham’s F12 حــاوی 5/2 % سرم جنین گاو و 15 % سرم اسب می‌باشد. زمان دو برابر شدن سلولها30 تا 52 ساعت است که به کیفیت سرم مورد استفاده بستگی دارد مطالعات نشان داده‌اند که اسید پکتیک یکی از عوامل افزایش دهنده مقدار ترشح پرولاکتین در این سلول‌ها در انکوباسیون‌های کوتاه مدت می‌باشد. انکوبه کردن سلولها با اسید پکتیک به مدت 30 دقیقه شکل سلولها را تغییر داده، سلول‌ها شروع به گرد و برجسته شدن کرده و حالت ترشحی به خود گرفتند. اسید پکتیک تاثیری در تعداد سلول‌ها نسبت به نمونه شاهد نداشت. محلول اسید پکتیک، محیط کشت را اسیدی می‌کند و باعث ایجاد شرایط نامطلوب برای سلول‌ها می‌شود. برای رفع این مشکل از محیط کشت حاوی Hepes به عنوان بافر برای ثبات pH استفاده شد ولی به مرور زمان شکل ظاهری سلول‌ها تغییر کرد و رشد آنها کاهش یافت. سلول‌ها گرد شدند، غشاء آنها گرانوله شد و درصد سلول‌های زنده کاهش پیدا کرد. این تغییر وضعیت سلول‌ها به دلیل تشکیل هیدروژن پر اکسید توسط Hepes می‌باشد. برای طبیعی ماندن مقدار pH محیط کشت و خنثی نمودن اسیدیته اسید پکتیک از محلول NaOH استفاده گردید که تاثیری بر سلول‌ها نداشت و بدینوسیله pH در حد 2/7 ثابت شد.}, keywords_fa = {پرولاکتین ‚ اسید پکتیک,صفت ذات}, url = {https://jos.ut.ac.ir/article_19488.html}, eprint = {https://jos.ut.ac.ir/article_19488_9b9c5a06098320ad201651db92e09be5.pdf} }
