An optimal increase of internal ABA concentration of leaves causes water saving through stomatal closure. In order to explore the effect of drought stress on leaf tissue ABA concentration and stomatal resistance of chickpea, a greenhouse experiment was conducted using five genotypes (Three tolerant genotypes including: ICCV93040, MCC13, MCC10 and two sensitive genotypes consisting of: MCC120 and MCC180) and two levels of water availability, growing at field capacity and at 25% of field capacity. Seeds were sown in 50 × 12 cm ‍ plastic tubes which were filled by washed sands. Nutrient elements were prepared using Hoagland nutrient solution. Leaf ABA concentration, determind using High Pressure Liquid Chromatography (HPLC). Results showed that drought stress imposed an increase in leaf ABA content up to flowering, but after this stage, leaf scenesence caused a reduction in leaf ABA. Genotypes showed different ABA concentrations and stomatal resistances in the presence of drought stress. The highest and lowest amount of ABA were observed in MCC13 (drought tolerant) and MCC120 (drought sensitive) respectively. ABA concentration of MCC13 and MCC120 were 29 and 3ngg-1 (fw) which is a 10 fold difference between them. Stomatal resistance of tolerant genotypes were more than sensitive ones, in sress and non-stress conditions. A significant positive correlation between leaf stomatal resistance and ABA concentration were observed (r = 0.87), but in non-strss conditions this relationship was not significant (r = 0.21).}, keywords = {Abscisic acid,stomatal resistance,Chickpea,Drought stress}, title_fa = {بررسی تغییرات آبسزیک اسید (ABA) بافت برگ و مقاومت روزنه‌ای در ژنوتیپ‌‌های مقاوم و حسّاس به خشکی نخود (Cicer arientinum L)}, abstract_fa = {آبسیزیک اسید (ABA) یک هورمون تنشی است که در سازگاری‌های گیاه به محیط‌های تحت تنش، دارای نقش‌های متفاوتی است. افزایش ABA بافت برگ، خروج آب از گیاه را از طریق بستن روزنه‌ها کاهش می‌دهد. به منظور بررسی تاثیر تنش خشکی بر تغییرات ABA بافت برگ و مقاومت روزنه‌ای نخود، آزمایشی در شرایط گلخانه انجام گرفت. سه ژنوتیپ مقاوم شامل ICCV93040، MCC13 و MCC10 و دو ژنوتیپ حساس به خشکی شامل MCC120و MCC180 در دو شرایط تنش خشکی و رطوبت کافی در لوله‌های پلاستیکی (cm12 × 50) حاوی شن شسته شده رشد نمودند و از محلول غذایی هوگلند برای تغذیه گیاهان استفاده ‌شد. میزان رطوبت شن، در تیمار شاهد در حد ظرفیت زراعی و در تیمار تنش 25% ظرفیت زراعی بود. غلظت ABA بافت برگ توسط دستگاه HPLC تعیین شد. تنش خشکی تاثیر معنی‌داری بر غلظت ABA بافت برگ داشت و با افزایش مدت زمان قرارگیری گیاه در معرض تنش تا زمان گل‌دهی غلظت ABA افزایش و پس از آن به دلیل پیر شدن برگها غلظت ABA کاهش یافت. ژنوتیپ‌های مورد بررسی از نظر تجمع ABA و مقاومت روزنه‌ای تفاوت نشان دادند. بیشترین و کمترین تجمع ABA به ترتیب در ژنوتیپ مقاوم به خشکی MCC13 و ژنوتیپ حساس به خشکی MCC120 با مقادیر(fw) ng g-13 و 29 مشاهده شد که یک اختلاف 10 برابری است. تنش خشکی مقاومت روزنه‌ای برگ را در ژنوتیپ‌های مقاوم و حساس به خشکی هر دو نسبت به تیمار شاهد افزایش داد. اما در هر دو تیمار مقاومت روزنه‌ای در ژنوتیپ‌های مقاوم بیش از ژنوتیپ‌های حساس بود. همبستگی معنی‌دار و مثبتی بین غلظت ABA و مقاومت روزنه‌ای برگ در شرایط تنش خشکی مشاهده شد (87/0 = r) اما در شرایط رطوبت کافی رابطه فوق معنی‌دار نبود (21/0 = r)}, keywords_fa = {آبسیزیک اسید,تنش خشکی,مقاومت روزآن‌ها ی,نخود}, url = {https://jos.ut.ac.ir/article_19489.html}, eprint = {https://jos.ut.ac.ir/article_19489_401912d722fe9e1b96a92503678cce29.pdf} }
